Mark Nawaqanitawase is leaving rugby league to pursue a Wallabies place at the upcoming World Cup, creating another wing vacancy for the Roosters.
Daniel Tupou's immediate retirement means coach Trent Robinson must find two new wingers for next season.
Tupou announced his decision days after helping the Roosters beat the Knights 19-18 in the 2026 NRL grand final.
The 35-year-old had been expected by some to play another season, but finished his career with four premierships.
He joined the Roosters in 2012 and leaves as the club's leading try-scorer, with 198 tries, and a one-club player.
"I've been thinking about this for a while, and I know the time is right for me to finish playing. It's hard to put into words what the Roosters have meant to me because this Club has been such a big part of my life," Tupou said in a statement.
"I feel incredibly blessed. I came here as a young kid just hoping for an opportunity and I could never have imagined the journey that would follow or everything I would be fortunate enough to experience along the way."
The club will also lose Angus Crichton to rugby union and Daly Cherry-Evans to Super League, where he is likely to end his career.
Connor Watson is heading to the Dragons for one season before joining PNG in 2028, leaving fans to anticipate a substantially changed Roosters lineup in 2027.
Tupou also thanked Robinson, his coach for almost his entire career.
"He's been my coach for almost my entire career and has had a huge influence on the player and the person I've become. He believed in me from the beginning, helped bring me out of my shell and has always challenged me to keep growing, not just as a footballer but as a man."
