England captain George Williams has failed in his second appeal against a two-match ban, meaning he will miss the first two Tests against Tonga.
The ban was initially imposed after Williams was found guilty of a shoulder charge in a play-off defeat by St Helens.
Despite his hopes of having the ban overturned, a new three-person tribunal upheld the suspension.
As a result, the 28-year-old will now be withdrawn from the England squad.
It is yet to be determined whether coach Shaun Wane will bring in a replacement for Williams or announce a new captain for the upcoming three-Test series starting on Sunday.
