O'Sullivan has more than 25 years' experience in recruitment and football, including roles with the Dolphins, New Zealand Warriors, Melbourne Storm and Sydney Roosters.
Brisbane appointed him General Manager of Recruitment after finishing 14th in its premiership-defence season.
"The Brisbane Broncos have appointed experienced recruitment specialist Peter O'Sullivan as the Club's new General Manager of Recruitment," the club's statement said.
His Knights contract reportedly ran until the end of 2029 at $350,000 per season. Reports put his Broncos deal at five years and $2.5 million, or $500,000 annually.
